Single family residences don't usually have to comply with ADA requirements.  That said, a clear space in a bathroom for wheel chair access should be at least 36"x48" and not be impeded by a door swing.  It should provide that clear space in front of all fixtures when the door is closed.

The general principle is to allow a wheelchair to enter the bathroom, close the door and manuever to all fixtures so as to use them.
